Bevacizumab (trade name Avastin, Genentech/Roche) is an angiogenesis inhibitor, a drug that slows the growth of new blood vessels. It is licensed to treat various cancers, including colorectal, lung, breast, glioblastoma, kidney and ovarian. It is also used off-label for the treatment of diseases of the eye in macular degeneration, diabetic retinopathy and retinopathy of prematurity.

Bevacizumab is given as an intravenous infusion every two to three weeks at the dose of either 7.5 mg/kg to 15 mg/kg. The intraocular dose is 1.25 mg to 2.5 mg in 100 ul (0.1ml) injections. The lower dose is usually preferred in premature newborns to prevent retinal blindness. The higher dose(s) is for adults with macular degeneration and severe forms of diabetic retinopathy given indefinitely at 4-6 weeks intervals.

The long-term local and systemic effect of Bevacizumab is unknown because of its long half-life and its suppression of VEGF (vascular endothelium growth factor). The Bevacizumab (Avastin) ELISA is a solid phase sandwich enzyme immunoassay. This assays incorporates a microtiter plate shaker in the first two steps of the assay protocol. Sample/calibrators are added to the microtiter plate wells that are coated with protein. Avastin is a whole antibody which binds, if present in the sample, to the protein coated microtiter plate wells. The contents in the wells following an incubation time are decanted or removed from the wells by washing. The enzyme conjugate is added to the wells. The enzyme conjugate is an anti-Avastin coupled to the enzyme Horseradish Peroxidase. Unbound enzyme conjugate is removed, after incubation. The wells are washed and a chromogenic substrate is added.
